My van came with a couple of block off plates for the hole on the drivers side (westy so the rear vents are covered up anyway.) and the PO duct taped the passenger side hole. Probably to keep the van warmer in the winter. Blocking that hole on the passenger and drivers side to keep the air flow from the rear does have a slight quieting effect as air is no longer flowing through the doors.
