Hydraulics should work immediately. If not, then you have something else going on. Tranny could be failing in some way. or shifter mechanism is not operating properly. I had the stiff shift problem on my 88 GL and solved it by changing from standard tranny oil to Redline MT 90. Made a huge difference. That might not resolve the issue with your tranny, but it is the cheapest check that I know of. If after a tranny oil change you still have the problems, the it's most likely overhaul time for the tranny.
