I tried it both ways.

Yes, the door was closed. In fact, I had done this switchover in my
garage the night before. I went out yesterday morning for an early
appointment, and tried to lock lock the car up. It was then that I
discovered that nothing I could do had any effect on the latch
mechanism. Upon removing the handle later in the day, I could see that
the keyed part works as it should. What doesn't work, apparently, is
the latch.
