With the cap off the drain tube could let air into the combustion chamber and possibly allow dangerous fumes to came out. I doubt that enough of either would flow to be an issue but I would not leave it off. The drain pipe tends to get partially or even completely plugged over the years, so just removing the cap may not let much water or gases flow. After the cap is off you can push a wire up into the pipe to see how blocked it is. Don't push anything in more than 12" . A bicycle spoke may work well.
