Sure it will work, except in those extreme circumstances where the engine has to be under a big load to open the leak. Those are hard to find. I have always done a similar test that doesn't use the glove (though I'd like to try it--strike that, I don't want a leaking head so I don't want to try it) but rather you just start the car from cold and run it about 30 seconds and shut it off. It doesn't have enough time to create pressure from normal engine heat, so if you remove the tank cap and hear a burp, you know you have a leak. Eventually that coolant is going to look dirty, anyway.
