Work fast! Seriously, clamp hose, remove from pump and place new hose on pump. Cut to length. With clamp in place on new hose quickly remove old and new one connected quickly. Have a bucket to catch what pours out. I loose than a quart.

How would one go about replacing that hose with fuel in the tank??

Can't clamp the hose - it has to come off. Figure there must be a procedure to get this change done and not make a complete firebomb waiting to be lit!
