That's the clamping screw that must be loosened first and tightened after.

That's the actual cam. It is a zinc casting and it has a hex recess in it which you will wallow out if you put force on it. Pull the plastic cover off, disassemble as needed and free up the cam where it's likely gotten frozen into the hole in the steel mounting plate.
